Well, one thing that might not be immediately obvious is that if you know the sum of the first three cells and the last three cells in a region, then you also know the sum of the middle three cells.
For instance, if you know that the sum of the first three cells is 22 and the last three cells is 14, then you know that the middle three cells in that region add up to 45 - (22 + 14) assuming we are talking about a 9x9 puzzle, so those middle three cells sum to 9.
Sometimes you will find it useful to work out what the sum of the middle three cells in a region are when solving outside puzzles, and this may even be required on occasion.
